  <common-name>Cobalt(II) succinate</common-name>
<description>Cobalt(II) succinate is an exogenous solid compound with the formula C4H4CoO4 and an average molecular weight of 175.01 g/mol. Cobalt(II) succinate belongs to the dicarboxylic acids and derivatives, a subclass of carboxylic acids and derivatives within the organic compounds.

The compound exhibits a wide range of biological interactions across 22 recorded protein targets. It inhibits heme synthesis by preventing the synthesis of 5-aminolaevulinate through the inhibition of 5-aminolevulinate synthase, both the erythroid-specific mitochondrial (ALAS2) and non-specific mitochondrial (ALAS1) forms. Additionally, cobalt inhibits carbonic anhydrases, specifically Carbonic anhydrase 1 (CA1), Carbonic anhydrase 2 (CA2), and Carbonic anhydrase 4 (CA4). It interferes with DNA repair by interacting with zinc finger DNA repair proteins, including Poly [ADP-ribose] polymerase 1 (PARP1). Furthermore, cobalt blocks high-voltage-activated calcium channels, which may impair neurotransmission; associated targets include the Voltage-dependent calcium channel gamma-1 subunit (CACNG1), Voltage-dependent calcium channel subunit alpha-2/delta-1 (CACNA2D1), Voltage-dependent calcium channel subunit alpha-2/delta-2 (CACNA2D2), Voltage-dependent calcium channel subunit alpha-2/delta-3 (CACNA2D3), and 10 other proteins.

Cobalt also interacts with specific antibodies, resulting in immunosensitization through interactions with Immunoglobulin heavy constant alpha 1 (IGHA1), Immunoglobulin heavy constant alpha 2 (IGHA2), and specific IgE antibodies. Finally, radioactive cobalt is capable of damaging lipids, RNA, and DNA through ionizing events.</description>

<mechanism-of-toxicity>Cobalt is believed to exhibit its toxicity through a oxidant-based and free radical-based processes. It produces oxygen radicals and may be oxidized to ionic cobalt, causing increased lipid peroxidation, DNA damage, and inducing certain enzymes that lead to cell apoptosis. Cobalt has also been shown to block inorganic calcium channels, possibly impairing neurotransmission. Cobalt can also chelate lipoic acids, impairing oxidation of pyruvate or fatty acids. In addition, cobalt may inhibit DNA repair by interacting with zinc finger DNA repair proteins, and has also been shown to inhibit heme synthesis and glucose metabolism. Cobalt may activate specific helper T-lymphocyte cells and interact directly with immunologic proteins, such as antibodies (IgA and IgE) or Fc receptors, resulting in immunosensitization. (L29)</mechanism-of-toxicity>
<metabolism>Cobalt is absorbed though the lungs, gastrointestinal tract, and skin. Since it is a component of the vitamin B12 (cyanocobalamin), it is distributed to most tissues of the body. It is transported in the blood, often bound to albumin, with the highest levels being found in the liver and kidney. Cobalt is excreted mainly in the urine and faeces. (L29)</metabolism>
