<common-name>Phalloin</common-name>

Phalloin is a solid exogenous compound with the chemical formula C35H48N8O10S. Phalloin belongs to the amino acids, peptides, and analogues, a subclass of carboxylic acids and derivatives within the organic compounds. With an average molecular weight of 772.87 g/mol, Phalloin is a heavy molecule.

This compound targets four proteins: cytoplasmic 1 actin (ACTB), cytoplasmic 2 actin (ACTG1), alpha skeletal muscle actin (ACTA1), and aortic smooth muscle actin (ACTA2). Phallotoxins bind actin at the interface between F-actin subunits, locking adjacent subunits together and preventing depolymerization, which poisons the cell. This mechanism leads to a decrease in the rate constant for the dissociation of actin subunits from filament ends, essentially stabilizing actin filaments. Additionally, these toxins inhibit the ATP hydrolysis activity of F-actin. Phalloin is one of a group of toxins known as phallotoxins isolated from the death cap (Amanita phalloides). The phallotoxins consist of at least seven compounds, all of which have seven similar peptide rings.

<mechanism-of-toxicity>Phallotoxins bind actin, preventing its depolymerization and poisoning the cell. They bind specifically at the interface between F-actin subunits, locking adjacent subunits together. This leads to a decrease in the rate constant for the dissociation of actin subunits from filament ends, which essentially stabilizes actin filaments through the prevention of filament depolymerization. Moreover, they inhibit the ATP hydrolysis activity of F-actin. (L1183, A2465)</mechanism-of-toxicity>
